When you jelq, do you release the first hand before you begin to move the 2nd hand up? Or do you hold the blood in the head at the top until the 2nd hand gets there?
I have been doing it the 2nd way and have these 2 red spots that won't go away. I think I have been doing it wrong, but I want to double check.

The 2nd way is fine and dont worry about the red spots. Its normal to get them and they will go away!

I usually do wet jelqs the second way, I think it is more intense. I will also do 50-100 wet jelqs were I kegel on each jelq before I grab the base with the second hand so more blood is in the shaft. It really ups the intensity, but since you have red spots I would take it a little easy. About a month ago I had a red spot that was about 1/8 in diameter. It didn't hurt or get bigger when I PE'd so I kept doing my normal workouts and it disappeared after 5 days or so.
